Software Module
AutoMorph
The AutoMorph module is used to automatically identify proximal and distal protoplasmic droplets and pig and bovine tail-coiled sperm. AutoMorph simultaneously integrate the analysis of vitality and density. The semen sample should be diluted with a clean diluent.
Protoplasmic droplets are produced during the maturation of a defective sperm. Defects can be caused by many factors, such as stress or disease.

Working principle of fluorescence analysis
The action is produced by a fluorescent stain with a wavelength. The structural integrity and functionality of sperm cells vary, and the coloration of fluorescent stains varies accordingly.

Analysis of the plasma membrane requires double staining, I .e. using Hoechst 33342/PI or SYBR14/PI.
The Hoechst 33342 stain penetrates the sperm cell membrane and is attached to its DNA. The sperm is stained blue. The PI stain (propidium iodide) can only penetrate into damaged sperm cells and can cover the Hoechst stain. Damaged sperm is stained red/purple. On this basis, AndroVision®The damaged and damaged sperm can be measured to calculate their proportion.
SYBR14/PI stain stains sperm green. PI penetrates into sperm with damaged cell membranes, which stains them red.
Analyzing the causes of viability

The acrosome analysis was performed using dual fluorescent staining H333342/FITC-PNA. Sperm are stained blue (H33342). Sperm with damaged acrosome are stained green (FITC-PNA).
